As long as your rotors are not directional from the factory, the rotation of the holes in the rotor does not matter. On higher-performance vehicles, those will generally be directional to increase the amount of airflow through them. Above all else, the most important feature of a rotor's function and cooling is instead the internal venting.
Most manufacturers will only drill one direction producing drilled or slotted versions of nondirectional rotors.

When looking at brake disks, there are a couple of things to note. The position (front left, rear right, etc) is usually found in the fitment information. On this BMW M5, the rotors are directional and are both noted as "front" and "left" or right". The left or right denotation is absent on non-directional rotors.
